5-AMINO-1MQ (5-amino-1-methylquinolinium) is a synthetic small-molecule inhibitor of nicotinamide N-methyltransferase (NNMT), an enzyme that consumes S-adenosylmethionine (SAM) and drains the NAD+ salvage pool in adipose tissue. Unlike peptide-based metabolic agents, it is a cell-permeable quinolinium compound that acts directly on the enzyme. Preclinical work by Neelakantan and colleagues (Biochemical Pharmacology, 2018) showed that NNMT inhibition reduced lipid accumulation in differentiating 3T3-L1 adipocytes, lowered white adipose mass and adipocyte size in diet-induced obese mice, and did so without reducing food intake.
Earlier work by Kraus and colleagues (Nature, 2014) established that NNMT knockdown protects against diet-induced obesity, providing the genetic rationale for pharmacological inhibition. Researchers use 5-AMINO-1MQ to model adipocyte metabolism, SAM/NAD+ balance, insulin sensitivity and energy expenditure, and to compare NNMT inhibition with incretin-based weight-loss pathways. The compound is supplied as 5mg of research-grade material in a 3ml vial. Store the powder at -20C, protect it from light, and dissolve it in an appropriate laboratory solvent such as DMSO or saline according to institutional protocols.
Common research questions concern its non-peptide classification, its selectivity for NNMT over related methyltransferases, and how its effects compare with GLP-1 agonists in diet-induced obesity models. This product is intended strictly for in-vitro and in-vivo laboratory research by qualified professionals. It is not approved for human consumption, clinical treatment or therapeutic use. A practical design consideration is solvent and route: 5-AMINO-1MQ is poorly suited to simple aqueous reconstitution and is typically prepared in DMSO or another laboratory solvent, then diluted into culture medium or vehicle for animal work.
Investigators comparing it with GLP-1 agonists should control for the different downstream pathways, because NNMT inhibition raises SAM and NAD+ availability rather than activating incretin receptors. Dose-response studies in diet-induced obese mice commonly run for several weeks and measure body composition, food intake, glucose tolerance and hepatic lipid content.
